Australia and Samoa have played each other five times at rugby union with Australia winning four times and Samoa once. Four of these test matches were played on Australian soil with the other played at a neutral venue as part of the 1991 Rugby World Cup. This is the only time the teams have played each other in a Rugby World Cup.

In 2011, Samoa was one of Australia's warm-up opponents ahead of the 2011 Rugby World Cup. Samoa was victorious, winning 32-23 in Sydney, the first time Manu Samoa had won a test match against Australia.
